Phil Burleigh, Chris Harris, Byron McGuigan, Nathan Fowles, Jamie Bhatti, George Turner, Darryl Marfo, Scott Cummings, Luke Hamilton and Jamie Ritchie are all the uncapped names in the squad that will face Samoa, New Zealand and Australia next month.
Full-back Stuart Hogg, who has only recently returned to action from a shoulder injury, has also been included along with fellow British & Irish Lion Tommy Seymour.
Townsend is looking forward to the three Test matches at Murrayfield and feels all the players deserve their chance with the national team as they build towards the 2018 Six Nations.
He told the SRU website: "The players selected have worked exceptionally hard at their clubs to earn inclusion in the national squad and we're looking forward to working with them throughout the autumn series.
"They will need to intensify their efforts further still to earn the opportunity and honor of play in front of capacity crowds in what are three exciting fixtures.
"We're facing two of the world's top three sides after a significant challenge from Samoa in our opening game. It will be an excellent challenge for our players."
